Peroxidase isoenzymes as markers for the rooting ability of easy-to-root and difficult-to-root Grevillea species and cultivars of Protea obstusifolia (Proteaceae)

摘要: One easy-to-root and one difficult-to-root species of the ornamental plant Grevillea were investigated for rooting potential in relation to peroxidase activity. In vitro-grown shoot segments both started root 30 d after transplanting medium containing indole-3-butyric acid (IBA), however, fewer roots found on G. petrophioides compared rondeau. Total (POX) activity was measured during process. showed higher total POX at time point adventitious formation than Isoelectric focusing electrophoresis that rondeau contained more acidic isoforms petrophioides, but basic prominent species, especially lateral emergence. addition, ability different hormones induced upper lower stem tested. Indole-3-acetic (IAA), IBA α-naphthaleneacetic rondeau, whereas same led induction petrophioides. Similar results obtained with Grevillea, variety Protea activity, middle part leaves. Feeding radiolabeled IAA resulted synthesis three compounds species. After 1h incubation no differences uptake appearance other labeled compounds. However, 2 4 h faster also metabolized a extent Three metabolites found, tentatively identified as IAA-aspartate, IBA, an conjugate.
