A Novel Model of Obesity-Related Diabetes: Introgression of the Leprfa Allele of the Zucker Fatty Rat into Nonobese Spontaneously Diabetic Torii (SDT) Rats

摘要: An fa allele of the leptin receptor gene (Leprfa) Zucker fatty rat was introduced into genome Spontaneously Diabetic Torii (SDT) rat, an inbred model nonobese type 2 diabetes mellitus, through `Speed congenic method'. The newly established strain a SDT for Leprfa maintained by intercrossing between fa-heterozygous littermates, and phenotypes related to obesity were investigated till 32 wks age. fa/fa rats both sexes exhibited obesity, adiposity insulin resistance associated with hyperphagia from loss action. Interestingly, they developed 5 age in males 8 females incidences reaching 100% at 16 73% females. In contrast, heterozygous (+/fa) wild-type (+/+) spontaneous approximately 20 wks, but not females, as original rats. These results indicate that accelerates onset making and/or potent risk factors development their diabetes. SDT.Leprfa is expected be novel obesity-related could useful tool studies genetic backgrounds response fa-induced obesity.
