摘要: Mexiletine is an antiarrhythmic agent used in the treatment of ventricular arrhythmia. The drug has a narrow therapeutic window which necessitates monitoring its serum concentrations. We describe gas chromatographic-mass spectrometric analysis mexiletine using selected ion monitoring. was extracted from alkaline with dichloromethane and then derivatized perfluorooctanoyl chloride. derivatization reaction completed 20 min at 80 degrees C. N-propylamphetamine as internal standard. ions monitored were m/z 122, 454 575 for 91, 118, 440 452 within-run precision concentration 1 mg/l 1.9% (mean=0.98, S.D.=0.019 mg/l, n=7) between-run 2.5% (mean=0.99, S.D.=0.025 n=7). assay linear concentrations 0.2 to 4 mg/l. detection limit 0.1 average recoveries standard 80% 84%, respectively There no carry over problem our assay. observed good correlation between measured by reference laboratory (GC) new GC-MS
