Identification of simple sequence repeats (SSRs) in olive ( Olea europaea L.)

摘要: A small insert genomic library of Olea europaea L., highly enriched in (GA/CT)n repeats, was obtained using the procedure Kandpal et al. (1994). The sequencing 103 clones randomly extracted from this allowed identification 56 unique inserts containing simple sequence repeat regions made by at least three single repeats. sample 20 primer pairs out 42 available were tested for functionality six olive varieties whose DNA served construction. All succeeded amplifying one product samples, and ten detecting more than allele used genetic characterisation a panel accessions belonging to 16 distinct varieties. total 57 alleles detected among genotypes polymorphic SSR loci. remaining pair amplification all plus longer fragment some genotypes. Considering polymorphism, 5.7 scored on average each dissimilarity matrix, based proportion shared pair-wise combinations genotypes, constructed disentangle relationships means UPGMA clustering algorithm. Graphical representation results showed presence two clusters first cluster grouped cultivated Ionian Sea coasts. second subdivisions: sub-cluster agglomerated inland areas Calabria; Basilicata Apulia nearby areas. Results analysis significant relationship between multilocus similarities geographic origin cultivars.
