摘要: Human envenoming by Lachesis muta venom, although infrequent, is rather severe, being characterized pronounced local tissue damage and systemic dysfunctions. Studies on the pharmacological actions of L. m. venom are relatively scant direct crude its purified phospholipase A(2) (PLA(2)) have not been addressed using in vitro models. In this work, we investigated cytotoxicity PLA(2) isoform LmTX-I cultured Madin-Darby canine kidney (MDCK) a skeletal muscle (C2C12) cell lines. As revealed neutral red dye uptake assay, (10 or 100 microg/ml) induced significant decrease viability MDCK cells. at concentrations tested (70-270 microg/ml 5-20 microM) displayed no both C2C12 Morphometric analysis Feulgen nuclear reaction increase chromatin condensation (pyknosis), apparent reduction number mitotic nuclei fragmentation some cells after incubation with venom. Monolayer exposure to resulted morphological changes as assessed scanning electron microscopy. The staining TRITC-labelled phalloidin showed marked disarray actin stress fiber following exposure. contrast, had effect nucleus morphologies well organization. These results indicate that exerts toxic effects probably does contribute per se cytotoxicity, these mediated metalloproteinases/disintegrins other components
