摘要: Abstract In this study, a signal peptide of AlkL was replaced with other peptides to improve the soluble expression and thereby facilitate transport dodecanoic acid methyl ester (DAME) substrate into E. coli. Consequently, FadL (AlkLf) showed higher activity toward DAME. Furthermore, promoter optimization for efficient heterologous transporter AlkLf alkane monooxygenase (AlkBGT) system conducted resulted in increased ω-oxygenation AlkBGT system. Moreover, bioinformatic studies led identification novel from Pseudomonas pelagia (Pel), which exhibited 20% towards DAME as compared AlkB. Finally, construction chimeric newly identified enabled production 44.8 ± 7.5 mM 12-hydroxy (HADME) 31.8 ± 1.7 mM dodecanedioic monomethyl (DDAME) two-phase reaction
