摘要: One of the unique features actinomycetes, especially genus Streptomyces, is presence linear plasmids. These range in size from 12 to 600 kb, and are often termed mega-plasmids. While many genes involved secondary metabolite production reside clusters on chromosome, several studies have identified biosynthetic large plasmids that produce important metabolites, including antibiotics. In this study, Pulse Field Gel Electrophoresis (PFGE) was used screen 176 actinomycete isolates for plasmids; these bacterial strains were previously isolated Great Salt Plains Oklahoma. Seventy-eight (44%) contained Several more than one plasmid, accounting a total 109 Ten showed extrachromosomal DNA larger 200 thus falling into category A subset 55 treated with S1 nuclease determine topology; all examined appeared be ranged ~55 400 kb. Eleven chosen Whole Genome Next Generation Sequencing. From 11 sequenced isolates, seven partially assembled. majority coded hypothetical proteins, others general functions, stress response, antibiotic heavy metal resistance. Draft genome sequences two mega-plasmid-bearing Streptomyces sp. strains, BF-3 4F, revealed production, antibiotic, resistance, osmoregulation, which likely facilitate their survival extreme halophilic environment. To our knowledge, first study explore harbored by actinomycetes
