摘要: Conditionally immortalized murine calvarial (CIMC) cells that support differentiation of precursors into mature osteoclasts were isolated. All six CIMC cell lines supported osteoclast in response to 1,25-dihydroxyvitamin D(3) or interleukin (IL)-11. CIMC-4 also tumor necrosis factor (TNF)-alpha, IL-1beta, IL-6. The resultant multinucleated expressed tartrate-resistant acid phosphatase and formed resorption lacunae on mineralized surfaces. cells, therefore, establish an assay is responsive many cytokines does not rely isolation primary stromal cells. Low concentrations the synergistically stimulated when cocultured with either Osteoclast induced by all stimuli other than TNF-alpha was completely blocked osteoprotegerin, whether stimulators examined alone combination. Moreover, study lack receptors showed induces primarily through direct actions precursors, which a distinct mechanism from used bone-resorptive agents this study.
