摘要: In Eukarya, the packaging of DNA into chromatin provides a barrier that allows for regulation access to genome. Chromatin is refractory processes acting on DNA. ATP-dependent remodeling machines and histone-modifying complexes can overcome this (or strengthen it in silencing processes). Both components (DNA histones) are subject postsynthetic covalent modifications, including methylation lysines (the focus chapter). These lysine marks generated by host histone methyltransferases (writers) be removed demethylases (erasers). Importantly, epigenetic modifications impact structure directly or read effector regulatory modules. Here, we summarize current knowledge structural functional properties various methyltransfereases demethylases, with emphasis their importance as druggable targets.
