T-cell receptor variable region genes in cutaneous T-cell lymphomas.

摘要: The genes encoding the T-cell receptor (TCR) variable beta (TCRBV) regions were studied in skin biopsy samples from 24 patients with cutaneous lymphomas (CTCL), i.e. mycosis fungoides (n = 7), Sezary syndrome 4), lymphomatoid papulosis 3) and large cell CTCL or without 7) CD30 expression. A panel of primers specific for TCRBV families 1-24 was designed applied to cDNA using a semiquantitative reverse transcriptase coupled polymerase chain reaction (PCR) method. Three showed restricted expression limited number (1-3) families. In remaining patients, an average 17 (range: 13-22) different expressed. All elevated (> 10%) individual significantly higher than that seen normal blood lymphocytes, but no preferential usage particular gene observed. Direct sequence analysis more 60 PCR products revealed clonal TCR transcripts 18 patients. single clone, constituting 9-100% (mean: 26%) mRNA, present 12 patients; two clones, 13-72% 21.5%) five three accounting < 0.5-13% one patient. six could not be identified. It is concluded most contain both non-clonal (reactive) T cells, latter cells are numerous anticipated. These may engaged tumour immune reactions, although prospective studies and/or serial investigations needed elucidate this issue fully.
