摘要: The epidermal growth factor receptor (EGFR) is a tyrosine kinase involved in cell that often misregulated cancer. Several recent studies highlight the unique structural mechanisms its regulation. Some elucidate important role juxtamembrane segment and transmembrane helix play stabilizing activating asymmetric dimer, suggest activation mechanism likely to be conserved among other human EGFR-related receptors. Other provide new explanations for two long observed, but poorly understood phenomena, apparent heterogeneity ligand binding formation of ligand-independent dimers. New insights into allosteric utilized by intracellular regulators EGFR hope sites could used as targets drug development.
