Morphological heterogeneity of HeLa cell mitochondria visualized by a modified diaminobenzidine staining technique.

摘要: The diaminobenzidine (DAB) technique for the ultrastructural localization of sites cytochrome c oxidase activity in animal tissues has been adapted to visualization mitochondria cells growing culture. modified allows staining all coverslip preparatins light microscopy. Electron microscopy thin sections material treated by this method revealed that mitochondrial profiles within a cell (and only these) are stained and they exhibit well preserved size internal structure. Coverslip cultures synchronized unsynchronized HeLa (F-315) with DAB reaction were examined under oil immersion. In majority cells, recognized as discrete bodies thinner peripheral portion cytoplasm. This observation indicates large proportion F-315 at least growth conditions used here, complement is dividied into distinct organelles. examination also considerable morphological heterogeneity mitochondria, which exhibited an ovoid or short rod-like or, less frequently, long filamentous shape, some evidence branching. variability morphology appeared be far more prounced between different than individual cells; cellular was not related any obvious way cell-cycle-dependent changes.
