摘要: Publisher Summary This chapter discusses the attempts to improve functional efficacy of fetal nigral transplants, focusing on three major questions. First, what extent is it possible use multiple intrastriatal grafts obtain a more complete reinnervation striatal complex in 6-hydroxydopamine (6-OHDA)-lesioned rat, and will such provide improvements over beyond that seen after standard limited transplants? Second, can dopamine (DA) neurone placed homotopically substantia nigra, induce recovery 6-OHDA lesion model, whether their axons be encouraged grow striatum? Third, inhibitory GABAergic neurons used restore function rat Parkinson model? Transplantation DA neurones into striatum rats with DA-denervating lesions fibre densities approximately 60%–70% normal levels throughout complex. Incomplete reported ectopic grafting. The results summarized suggest intranigral grafting GABA-rich tissue some drug-induced rotational behavior skilled forelimb use. GABA released within output nuclei may tonic inhibition outflow, similar obtained by high-frequency deep brain stimulation, thus offer new route for therapeutic intervention Parkinson's disease.
