Characteristics and prevalence within serogroup O4 of a J96-like clonal group of uropathogenic Escherichia coli O4:H5 containing the class I and class III alleles of papG

摘要: The recent discovery of a geographically dispersed clonal group Escherichia coli O4:H5 that includes prototypic uropathogenic strain J96 prompted us to determine the prevalence J96-like strains within serogroup O4 and further assess characteristics such strains. We used O:K:H;F serotyping, PCR-based genomic fingerprinting, pulsed-field gel electrophoresis (PFGE), multilocus enzyme (MLEE), PCR detection three papG alleles cytotoxic necrotizing factor 1 (cnf1) aerobactin (aer) gene sequences characterize 15 among 336 E. isolates from clinical collections (187 mixed-source bacteremia, 75 urosepsis, 74 acute cystitis). constituted approximately half strains, or 2% total population. In contrast other characteristically exhibited specific III capsular antigens, H5 flagellar F13 fimbrial distinctive fingerprint, class allele (plus, in 50% enigmatic I allele), cnf1 but lacked aer. A subset these was remarkably homogeneous with respect all PFGE fingerprint MLEE pattern. These findings clarify epidemiological relevance as model extraintestinal pathogen, provide evidence outside J96, offer insights into evolution O4.
