Inhibition of Aldosterone Production by Adrenomedullin, a Hypotensive Peptide, in the Rat

摘要: Recently, we conducted in vitro studies and reported that adrenomedullin, a novel hypotensive peptide, inhibits aldosterone secretion by dispersed rat adrenal zona glomerulosa cells. To assess the physiological role of this inhibitory effect, investigated effect adrenomedullin on production vivo. Male Sprague-Dawley rats were fed normal sodium diet before experiments. begin experimental procedure, stimulated with sodium-deficient or bilateral nephrectomy. After 3 days depletion immediately after nephrectomy, injected synthetic human (2.5 nmol/kg SC) repeated injection three times at 6-hour intervals. Two hours last injection, decapitated capsular tissue was collected. Adrenomedullin had no plasma concentrations diet. Rats significantly increased both (4770.1±364.3 pmol/L) gland (57.34±3.27 pmol per adrenal). Subsequently, inhibited increases (plasma, 2648.9±313.2 pmol/L; adrenal, 44.28±4.94 In nephrectomized rats, also adrenomedullin. second part study, renin concentration, activity, corticosterone serum potassium immunoreactive concentration examined for effects. The first four unaffected, last, elevated 15% to 30%. These vivo results, together our data, suggest may indeed play control blood pressure electrolyte balance.
