Oral supplementation of silibinin prevents colon carcinogenesis in a long term preclinical model.

摘要: Chemoprevention through dietary intervention is an emerging option to reduce colon cancer mortality. β-catenin plays important role in the Wnt signaling cascade that most commonly dysregulated colorectal cancer. Our aim was explore modulatory effect of silibinin on expression employing 1,2-dimethylhydrazine (DMH) induced male Wistar rats as experimental model during different stages carcinogenesis. Colon tissues were analyzed for β-catenin, proliferating cell nuclear antigen (PCNA) and argyrophilic nucleolar organizer regions by using immunohistochemistry silver staining. Immunoblotting employed study cyclin D1 expression. Glutathione (GSH) glutathione related enzymes assayed spectrophotometric analysis. Silibinin inhibited DMH-induced decreasing tumor incidence multiplicity. supplementation DMH-treated restored levels GSH-dependent decreased PCNA, D1. Mechanistically inhibits carcinogenesis modulating Wnt/β-catenin pathway redox system. Since highly sensitive adults who may have preneoplastic lesions their be benefited silibinin.
