Axonal transport and localization of B-50/GAP-43-like immunoreactivity in regenerating sciatic and facial nerves of the rat.

摘要: Neurons that can regenerate their axons following axotomy increase synthesis and axonal transport of a growth-associated protein, called GAP-43, which has been shown to be identical the synaptic phosphoprotein B-50. The function B-50/GAP-43 process regeneration is unknown. We used polyclonal, affinity-purified antibody against B-50 study localization B-50/GAP-43-like immunoreactivity (B50LI) in regenerating sciatic facial nerves adult rats. Quantitative data were obtained by densitometry band immunoblots nerve segments, had run on SDS-polyacrylamide gels. In nerve, anterograde accumulation at collection ligature was 3.0 times higher than retrograde accumulation. mobile fraction B50LI only 0.28 total traveled with mean velocity 5.3 mm/hr. distribution newly regenerated portion revealed maximal levels midway between position crush fastest-growing axons. Immunocytochemistry this demonstrated associated but also large extent extra-axonal structures outlining Schwann cell bands Bungner. This zone B50LI-positive found extend more distally processed longer, e.g., up 5 mm distal after 3 d 8 4 d. Further zone, many fine profiles could detected antibody, neurofilament negative. These findings raise possibility an B-50/GAP-43, as protein might secreted from play role axon-Schwann interactions during maturation.
