摘要: Immunohistochemical localization of choline acetyltransferase (ChAT) in cholinergic neurons has been difficult to achieve because problems encountered producing specific antisera. Here we describe the production and characterization several distinct monoclonal antibodies ChAT. Each exhibits one three general patterns cross-species reactions; pattern shows reactivity limited mainly bovine ChAT, a second only ChAT from higher mammals including humans, third all tested. The bound specifically two closely related proteins 68,000 70,000 daltons using Western blotting technique. One was used localize immunohistochemically known structures rat brain, motor neurons, basal forebrain neostriatal neurons.
