Liver X receptor reduces proliferation of human oral cancer cells by promoting cholesterol efflux via up-regulation of ABCA1 expression

摘要: Liver X receptors (LXRs) contribute not only to maintain cholesterol homeostasis but also control cell growth. However, the molecular mechanisms behind LXR-mediated anti-proliferative effects are largely unknown. Here we show, by immunohistochemistry, that LXRα and LXRβ differentially distributed in oral stratified squamous epithelia. By immunohistochemical Western blot analyses, reveal is abundantly expressed human carcinoma (HOSCC) tissues lines. Cell counting, BrdU labeling cycle assay indicated LXR stimulation led significant reduction of proliferation HOSCC cells. Importantly, our study highlights, using RNA interference, ATP-binding cassette transporter A1 (ABCA1)-accelerated efflux critical for growth inhibitory action LXRs Moreover, demonstrate activation reduces xenograft tumour cells mice accompanied upregulation ABCA1 expression decline levels tumour. These findings strongly suggested targeting LXR-regulated transport, yielding lowering intracellular levels, could be a promising therapeutic option certain types cancers.
