摘要: Activation of the protein kinase A (PKA) signaling system is necessary for FSH-induced granulosa cell differentiation, but it not known whether activation PKA sufficient to account complex pattern gene expression that occurs during this process. We addressed question by infecting cells with a lentiviral vector directs constitutively active mutant (PKA-CQR) and compared cellular responses PKA-CQR stimulated FSH. Expression in undifferentiated resulted induction both estrogen progesterone production absence cAMP. The stimulatory effects FSH on were suppressed inhibitor H-89 mimicked PKA-selective cAMP agonists. mRNA levels P450scc 3β-HSD induced similar extent PKA-CQR, whereas P450arom LHr greater Microarray analysis profiles revealed majority genes appeared be comparably regulated some appear than PKA-CQR. These results indicate pathway most (as identified microarray analysis), including those biosynthetic differentiation. However, optimal aromatase, LHr, other appears require additional pathways.
