摘要: Human embryonic stem cells are a promising tool to study events associated with the earliest ontogenetic stages of hematopoiesis. We describe generation erythroid from hES (H1) by subsequent processing present at early and late embryoid body (EB) differentiation. Kinetics hematopoietic marker emergence suggest that CD45+ hematopoiesis peaks D14EB differentiation stages, although low-level CD45- can be seen before stage. By morphologic criteria, hES-derived were definitive type, but these both mRNA protein levels coexpressed high (ϵ) fetal (γ) globins, little or no adult globin (β). This expression pattern was not altered presence absence bovine serum, vascular endothelial growth factor, Flt3-L, coculture OP-9 during culture time dependent. The coexpression globins definitive-type does faithfully mimic either yolk sac their liver counterparts.
