The differential effects of 2% oxygen preconditioning on the subsequent differentiation of mouse and human pluripotent stem cells

摘要: A major challenge facing the development of effective cell therapies is efficient differentiation pluripotent stem cells (PSCs) into pure populations. Lowering oxygen tension to physiological levels can affect both expansion and stages. However, date, there are no studies investigating knock-on effect culturing PSCs under low conditions on subsequent lineage commitment at ambient levels. were passaged three times 2% O2 before allowing spontaneously differentiate as embryoid bodies (EBs) in high (20% O2) conditions. Maintenance mouse was associated with a significant increase expression early markers FGF5 Eomes, while conversely we observed decreased these genes human PSCs.
