Effect of estradiol on fibroblasts from postmenopausal idiopathic carpal tunnel syndrome patients.

摘要: Fibrosis of the subsynovial connective tissue (SSCT) is a characteristic finding in patients with idiopathic carpal tunnel syndrome (CTS). Idiopathic CTS frequently occurs postmenopausal women; therefore, female steroid hormones, especially estrogens, may be involved its development. In this study, we evaluated effect estradiol on expression genes and proteins related to fibrosis SSCT fibroblasts from CTS. This study included 10 women (mean age 76 years). Fibroblasts derived were treated (10-4 -10-12  M), levels TGF-β-responsive evaluated. The relationships between untreated estrogen receptor α (ERα) ERβ changes gene due treatment examined by quantitative real-time polymerase chain reaction. effects 10-4  M collagen type I (Col1) III (Col3) protein also fluorescent staining. ERα/β Col1/3 immunohistochemical reduction Col1A1 mRNA was positively correlated ERα (r = 0.903, p < 0.01). At level, Col1 Col3 down-regulated. These results indicated that ERα-mediated signaling regulation Col1A1, regulatory dependent level. accurate evaluation level individual might guide effective use new replacement therapy.
