摘要: Abstract Inflammation response and oxidative stress play important roles in acute lung injury (ALI). Activation of the cAMP/protein kinase A (PKA) signaling pathway may attenuate ALI by suppressing immune responses inhibiting generation reactive oxygen species (ROS). Hydroxysafflor yellow (HSYA) is a natural flavonoid compound that reduces inflammatory cytokine-mediated damage. In this study, we examined whether HSYA could protect lungs from oleic acid (OA)-induced injury, which was used to mimic ALI, determined role cAMP/PKA process. Arterial tension (PaO 2 ), carbon dioxide tension, pH, PaO /fraction inspired ratio blood were detected using gas analyzer. We measured wet/dry weight evaluated tissue morphology. The protein cytokine levels bronchoalveolar lavage fluid serum enzyme-linked immunoassay. activities superoxide dismutase, glutathione peroxidase, PKA, nicotinamide adenine dinucleotide phosphate oxidase, concentrations cAMP malondialdehyde assay kits. Bcl-2, Bax, caspase 3, p22 phox analyzed Western blotting. OA increased ROS caused dysfunction decreasing synthesis, PKA activity, stimulating reducing Bcl-2/Bax ratio. H-89 these effects. significantly antioxidant enzymes, inhibited via activation, attenuated OA-induced injury. Our results show required for protective effect against ALI.
