A Retroviral Wild-type p53 Expression Vector Penetrates Human Lung Cancer Spheroids and Inhibits Growth by Inducing Apoptosis

摘要: Abstract Multicellular tumor spheroids approximate the three-dimensional configuration of primary and metastatic tumors. The effects retrovirus-mediated transduction wild-type p53 (wt- ) were studied on multi-cellular human non-small cell lung cancer lines H322a, gene which is homozygously mutated at codon 248, WT226b, has endogenous wt- . growth WT226b was not affected by exogenous transduction; H322a spheroids, however, significantly inhibited addition virus stocks. Transduction cells retroviral vector penetration multiple layers in demonstrated situ polymerase chain reaction/hybridization with neomycin-resistant neo probe. Apoptotic changes indicating programmed death observed treated virus. These results suggest that vectors can penetrate into masses induce potentially therapeutic effects.
