Core–shell microspheres delivering FGF-2 and BMP-2 in different release patterns for bone regeneration

摘要: Bone regeneration by applying fibroblast growth factor-2 (FGF-2) plus bone morphogenetic protein-2 (BMP-2) is considered advantageous over a single factor (GF) treatment because the spontaneous repair of defects essentially regulated series GFs, including FGF-2 and BMP-2. However, temporal interactions between BMP-2 remain elusive how to take full advantage bottleneck in translating this dual-GF strategy into clinical applications. To compare long-term effects different patterns on regeneration, novel delivery system needed. Herein, we report type PLLA core–PLGA shell double-walled microsphere. Different release core shell, respectively, were achieved due distributions these two GFs. In vivo evaluations dual GFs using rat graft model suggested that sequential helped bridge remodel critical-sized grafts more efficiently than other patterns. More importantly, core–shell microspheres simultaneously continuously releasing resulted obvious resorption grafted non-union at 4 weeks. Consistently, vitro bovine sections led enhanced osteoclastogenesis compared GF treatments. The organization taking will enable development efficient devices for existing systems.
