摘要: Porphyrins and metalloporphyrins are strong DNA binders. Some of these compounds have been used for radiation sensitization therapy cancer targeted to interact with cellular DNA. This study was designed examine the interaction calf thymus chlorophyll a (CHL) in aqueous solution at physiological pH CHL/DNA(phosphate) ratios (r) 1/160, 1/80, 1/40, 1/20, 1/10, 1/5. Fourier transform infrared (FTIR) difference spectroscopy characterize nature DNA-pigment interactions establish correlations between spectral changes CHL binding mode, constant, sequence selectivity, secondary structure, structural variations DNA-CHL complexes solution. Spectroscopic results showed that is an external binder no affinity intercalation. At low pigment concentration (r = 1/40), there two major sites on duplex: 1) Mg-PO2 2) Mg-N7 (guanine) overall constant K 1.13 x 10(4) M-1. The distributions 60% backbone PO2 group 20% G-C base pairs. associated reduction B-DNA structure favor A-DNA. high content 1/10), helix opening occurs, alterations A-T bases. (1/5), aggregation observed, which does not CHL-DNA complexation.
