摘要: Aim: To investigate the effect of antisense compounds (AS) targeting human p53 mRNA on radiosensitivity MCF-7 cells. Methods: Western blotting and RT-PCR were used to analyze protein content level. Additionally, cell proliferation, cycle apoptosis all analyzed in irradiated or sham-irradiated Results: Among five (AS), AS3 was identified efficiently inhibit level content. Interestingly, transfer has little proliferation DU-145 cells (mutant p53) after ionizing radiation (IR). In contrast, a marked increase growth inhibition observed (wild-type p53), suggesting that can cells. it also transfection decreased fraction G 1 phase cells, increased proportion S compared untreated 24 h IR lines. Conclusion: increases induced by 5 Gy-radiation, this mechanism maybe closely associated with abrogation arrest.
