Characteristics of a negative thyroid hormone response element.

摘要: Thyroid hormones may stimulate or repress transcriptional activity depending upon the specific gene. Whereas, a palindromic DNA sequence, TREpal, mediates positive regulation by thyroid hormone, negative response element (nT3RE) remains undefined. Therefore, we have examined sequences that mediate inhibitory effects of hormone on transcription beta-subunit gene rat thyrotropin (rTSH beta). In pituitary tumor cells (GH3), transient expression plasmid constructs containing putative nT3RE rTSH beta mediated L-triiodothyronine (T3). Since this contained which resembled half-site motif consensus T3RE and idealized palindrome (TREpal), tested construct in same cells. T3 decreased plasmid. Cotransfection studies T3-receptor (T3R)-deficient indicated either alpha isoforms T3R were required for hormone. Both isomers bind to from TREpal. summary, our results show repressive properties are beta. Unexpectedly, resembles component TREpal enhances promoter T3.
