摘要: The majority of tumors express an isoform the glycolytic enzyme pyruvate kinase, type tumor M2. isoenzyme exists in active tetrameric and a less dimeric form. form is strongly overexpressed cells this new marker thus called M2-PK. This released from quantitatively detectable body fluids by sensitive enzyme-linked immunosorbent assay (ELISA). To establish ELISA for routine diagnostic clinical laboratory, sample stability was evaluated. Therefore, blood samples were collected different ways healthy donors. Reproducibility M2-PK detection EDTA-plasma not affected day to 'stress' (e.g. shaking, leaving at room temperature several hours without prior centrifugation). Similar results obtained with citrate-plasma, whereas serum heparin-plasma only reproducible when centrifuged within 2 hrs after collection. It appears that lymphocytes contain small amounts isoenzyme. They might release samples, but samples. indicated be most appropriate as marker.
