Formalin-fixed, paraffin-embedded tissues sectioned at 5 lm stained either hematoxylin eosin (HE) or Congo red without KMnO, pretreatment. red-stained sections examined both bright field polarized light microscopy. Using modified avidin-biotin-peroxidase complex (ABC) method,> evaluated for guinea pig anti-porcine serum (Dako Corp., Carpinteria, CA) diluted 1 : 800 phosphate-buffered saline (PBS), pH 7.4; 400 rabbit anti-human glucagon (Dako); 100 somatostatin calcitonin gene-related peptide (CGRP) (Peninsula Laboratories, Belmont, CA); 200 amylin (Peninsula). Positive controls included internal tissue containing application primary antisera normal bonnet radiata) pancreas. Negative replacing appropriate nonimmune sera applying canine kidney glomerular amyloid. Five-micrometer collected on positively charged glass slides (Fisher Scientific, Pittsburgh, PA), deparaffinized, rehydrated, incubated 1.5% goat PBS 20 minutes room temperature humidified chamber.
