摘要: The causal link between uncontrolled Hedgehog (Hh) pathway activation and oncogenesis has inspired the development of small-molecule inhibitors this developmental signaling pathway. In chapter, we discuss how a cyclopia-inducing natural product isolated in 1950s engendered pharmacopoeia compounds that block Hh target gene expression. We describe these were discovered, their mechanisms action, potential as anti-cancer agents. also examine limitations current approaches future directions for pathway-targeting chemotherapies.
