Involvement of hydrogen peroxide in asbestos-induced NFAT activation.

摘要: The present study investigated the role of reactive oxygen species (ROS) in activation nuclear factor activated T cells (NFAT), a pivotal transcription responsible for regulation cytokines, by asbestos mouse embryo fibroblast PW cells. Exposure to led transactivation NFAT time- and dose-dependent manner. Scavenging asbestos-induced H2O2 with N-acety-L-cyteine (NAC, general antioxidant) or catalase (a specific inhibitor) resulted inhibition activation. In contrast, an increase generation addition superoxide dismutase (SOD) slightly enhanced addition, pretreatment sodium formate did not exhibit any activity induced asbestos. These results demonstrated that appeared play important transactivation. Furthermore, it was observed incubation 12-O-tetradecanoylphorbol-13-acetate (TPA) only itself, but also induction. Pretreatment cyclosporin A (CSA), pharmacological inhibitor phosphatase calcineurin, blocked both asbestos- TPA plus data suggest is able induce through H2O2-dependent CSA-sensitive pathways, which may be involved carcinogenesis.
