摘要: A number of phosphoinositide-specific phospholipases C (PLC) different species have recently been cloned. The predicted amino acid sequences these isoforms contain two highly conserved domains. Here we report the identification a PLC gene Dictyostelium by using polymerase chain reaction. Primers were designed coding for regions located within one domains PLCs. Cloning and sequencing reaction product revealed unique PLC-like sequence. This sequence was used to screen library isolate several overlapping cDNA clones. complete expressed in cells resulting increased basal levels inositol 1,4,5-trisphosphate enhanced activity. identified PLC, DdPLC, encodes protein with calculated molecular mass 91 kDa. deduced contains found other isoforms, separated short variable region. C-terminal part shows strong homology mammalian PLC-delta isoform. DdPLC is at all stages development, an increase transcription during starvation culminating fruiting body.
