摘要: Expression of the src gene Rous sarcoma virus (RSV) in chicken embryo neuroretinal (NR) cells results morphological transformation and sustained proliferation a normally resting cell population. We have previously reported isolation mutants RSV which retain full growth-promoting activity while displaying reduced transforming properties. Two such mutants, PA101 PA104, were used to investigate whether p60src-associated kinase is required for mitogenic function src. A comparison patterns phosphorylation wild-type mutant p60src revealed that tyrosine residues PA104 was markedly reduced, whereas relative amount phosphotyrosine comparable protein. In vitro immunoprecipitated from NR infected with or as measured by heavy chains specific immunoglobulin G molecules 1/10 molecule. Moreover, when temperature sensitive capacity maintained at either permissive restrictive growth, quantitation indicated could not be linked absolute level p60src. Transformation resulted 10-fold increase total cellular 34K protein, possible vivo substrate contrast, targets PA104. These indicate does require elevated levels activity.
