摘要: Emerging evidence indicates that metabotropic glutamate receptor 5 (mGluR5) critically modulates drug and drug-related behaviors. However, the role of mGluR5 in opiate-induced contextual memory remains unclear. Here, we found microinfusion antagonist 3-((2-Methyl-1,3-thiazol-4-yl)ethynyl)pyridine (MTEP) into nucleus accumbens (NAc) shell, but not core, significantly attenuated expression morphine conditioned place preference (CPP) rats. Following CPP, protein level membrane was selectively increased NAc shell. In primary striatal neurons, observed treatment with agonist CHPG phosphorylation extracellular signal-regulated kinase (ERK), which dependent on mGluR5-inositol-1,4,5-trisphosphate-reactive oxygen species (ROS) pathway. Moreover, microinjection ROS scavenger Tempol shell rats blocked CPP. Further, administration t-BOOH, a donor, rescued retrieval impairment CPP produced by MTEP. Our previous study demonstrated ERK Thus, results present suggest is essential for memory, mediated at least part, through ROS/ERK signaling Uncovering molecular basis opiate will benefit development new therapeutic approaches addiction.
