Natural history of hepatic metastases from colorectal cancer - pathobiological pathways with clinical significance

摘要: Colorectal cancer hepatic metastases represent the final stage of a multi-step biological process. This process starts with series mutations in colonic epithelial cells, continues their detachment from large intestine, dissemination through blood and/or lymphatic circulation, attachment to sinusoids and interactions sinusoidal such as endothelial Kupffer stellate cells pit cells. The metastatic sequence terminates colorectal cell invasion, adaptation colonisation parenchyma. All these events, termed invasion-metastasis cascade, include multiple molecular pathways, intercellular expression plethora chemokines growth factors, adhesion molecules, selectins, integrins or cadherins, well enzymes including matrix metalloproteinases. review aims present recent advances that provide insights into cell-biological events emphasizes those may be amenable therapeutic targeting.
