Infrared imaging of MDA-MB-231 breast cancer cell line phenotypes in 2D and 3D cultures

摘要: One current challenge in the field of breast cancer infrared imaging is identification carcinoma cell subtypes tissue. Neither sequencing nor immunochemistry currently able to provide a by thorough classification. The latter needed build accurate statistical models capable recognizing diversity lines that may be present tissue section. possible approach for overcoming this problem obtain IR spectral signature well-characterized tumor culture. Cultures three-dimensional matrices appear generate an environment mimics better vivo environment. There are, at present, series have been thoroughly characterized two- and (2D 3D) cultures full transcriptomics analyses. In work, we describe methods used grow, process, characterize triple-negative line, MDA-MB-231, 3D laminin-rich extracellular matrix (lrECM) culture compare it with traditional monolayer sections. While unsupervised analyses did not completely separate spectra cells grown 2D from lrECM cultures, supervised analysis resulted almost perfect separation. When responses epithelial clinical samples were added these data, principal component indicated they cluster closer than on flat plastic substrata. This result encouraging because correlating line features biopsies.
