摘要: Today, proteomics usually compares clinical samples by use of bottom-up profiling with highresolution mass spectrometry, where all protein products a single gene are considered asan integral whole. At the same time, proteoforms, which considers varietyof species, offers potential to discover valuable biomarkers. Proteoforms areprotein species that arise as consequence genetic polymorphisms, alternative splicing,post-translational modifications and other less-explored molecular events. The comprehensiveobservation proteoforms has been an exclusive privilege top-down proteomics. Here,we review possibilities approach address microheterogeneity thehuman proteome. Special focus is given shotgun structure-basedbioinformatics source hypothetical can potentially be verified bytargeted spectrometry determine relevance diseases.
