Photodynamic therapy of tumours and other diseases using porphyrins

摘要: Photodynamic therapy (PDT) with porphyrins and red light (620–630 nm) is finding increasing clinical application for both the eradication of relatively small tumours palliation inoperable or obstructive tumours. PDT also shows some promise sterilization tumour bed after surgical removal neoplastic masses. Several have been found to be accumulated retained by tissues; however, a chemically prepared derivative haematoporphyrin, termed HpD, purified form DHE (dihaematoporphyrin ether ester), are most frequently used in practice owing their optimal tumour-localizing properties low systemic toxicity dark. The efficiency HpD/DHE photoactivation very low, since extinction coefficient at wavelengths above 600 nm below 103 m −1 cm−1. Therefore, large number investigations being performed order improve efficacy PDT. One approach involves use porphyrin analogs (e.g., chlorins, phthalocyanines) which retain high affinity possess intense absorption bands spectral region. Moreover, selectivity targeting can enhanced transport photosensitizing drug types lipoproteins monoclonal antibodies. These developments interest view proposed extension treatment other diseases, including viral microbial infections, atheroma psoriasis.
