摘要: MicroRNAs (miRNA) comprise a group of short ribonucleic acid molecules implicated in regulation key biological processes and functions at the post-transcriptional level. Ionizing radiation (IR) causes DNA damage generally triggers cellular stress response. However, role miRNAs IR-induced response human embryonic stem cells (hESC) has not been defined yet. Here, by using system biology approaches, we show for first time, that miRNAome undergoes global alterations hESC (H1 H9 lines) after IR. Interrogation expression levels 1,090 miRNA species irradiated showed statistically significant changes 54 genes following 1 Gy X-ray exposures; were found to be highly temporally cell line - dependent hESC. Time-course studies 16 hr is much more robust compared 2 hr-response signature (only eight genes), may involved regulating cycle. Quantitative real-time PCR performed on some confirms robustness our microarray platform. Positive differentiation-, cycle-, ion transport- endomembrane system-related predicted negatively affected Our findings reveal fundamental modulating response, identify novel molecular targets
