摘要: Tissue factor (TF) has been implicated in the thrombotic complications seen during vascular rejection of allografts and may contribute to intimal hyperplasia chronic allograft vasculopathy. Downregulation endothelial TF expression post-transplantation could therefore be therapeutic value. Lentivirus-mediated RNA interference was used primary cells (EC) investigate its effects on protein functional activity. a TF-specific short-interfering (si) with green fluorescent as reporter gene (siRNATF-GFP) resulted 42 ± 3.9% reduction EC surface-expressed compared expressing scrambled siRNATF sequence (P = 0.025). The content lysates reduced from 6.85 ± 1.99 ng 3.05 ± 0.82 ng (P = 0.006). Factor X (FX) activation not impaired apical surface. subendothelial matrix ECs low showed significantly activity non-transduced or harboring empty vector. siRNATF-GFP exhibited (GFP) cell density an altered morphology. Transfection control high (J82 cells) (MiaPaCa-2 oligonucleotides caused apoptosis J82 but MiaPaCa-2 cells. Thus, lentivirus-mediated reduces activated does affect FX by TF/FVIIa expressed downregulation nevertheless substantial negative viability TF-expressing These findings imply that certain levels are required for maintained growth endothelium tumor
