摘要: The S49 mouse lymphoma mutant cell line Kin- is resistant to the cytotoxic effects of elevated cAMP levels, has no detectable cAMP-dependent protein kinase activity, and depressed levels cAMP-binding regulatory subunits. We demonstrate that although lacks catalytic subunit protein, these cells express wild-type mRNA for both C alpha beta isoforms. Translation appears be normal in cell, based on observation associates with large polyribosomes cells. cloned cDNA from show its transient expression another type leads activation a cAMP-sensitive luciferase reporter gene, suggesting functional made. In addition having inhibited presence RI subunit, indicating formation holoenzyme complex normal. suggest mutation responsible phenotype cellular component directly or indirectly causes degraded rapidly.
