摘要: To fully exploit the transfection technology developed for Plasmodium we investigated features of replication, expression and segregation an episomally maintained DNA construct during asexual blood stage development in genetically transformed parasites P. berghei. Using situ hybridisation techniques were able to show that introduced is located nucleus parasite not segregating uniformly schizogony. Replication mainly takes place between 16 24 h after invasion merozoites, coinciding with chromosomal replication. Furthermore plasmid-borne DHFR/TS gene constitutively transcribed throughout development.
