In vivo structural characterization of the whole SARS-CoV-2 RNA genome identifies host cell target proteins vulnerable to re-purposed drugs

摘要: SARS-CoV-2 is an RNA virus of the Coronaviridae family that causal pathogen ongoing Coronavirus Disease 2019 pandemic. There are currently no antiviral drugs or vaccines to treat COVID-19, and failure identify effective interventions can be blamed on our incomplete understanding nature this its host cell infection process. Here, we experimentally determined structural maps genome in infected human cells also characterized vitro refolded structures for 6 other coronaviruses. Our vivo data confirms several elements predicted from theoretical analysis goes much further revealing many previously unknown features functionally impact viral translation discontinuous transcription cells. Importantly, harnessed structure alongside a deep-learning tool accurately dozen related proteins bind genome, none which were known previously. Thus, study lays foundation coronavirus biology indicates promising directions rapid development therapeutics COVID-19.
