摘要: Role of endothelin in cyclosporine-induced glomerular dysfunction. Since recent studies indicate that cyclosporine (CsA) disrupts endothelial integrity and injured cells release excess endothelin, we examined endothelin's role acute nephrotoxicity. Following CsA (20 mg/kg i.v.), rabbit anti-porcine (aE) serum was continuously infused into a first order branch the main renal artery Munich-Wistar rats whereupon hemodynamics glomeruli not with aE as well those within same kidney were simultaneously assessed by micropuncture techniques. In treated kidneys, aE, single nephron GFR (SNGFR) plasma flow rate (Q A ) fell profoundly (on average 42 48%, respectively) below baseline values association lower capillary pressure elevated afferent arteriolar resistance. By contrast, this vasoconstrictive pattern markedly attenuated: SNGFR was, on average, only 19% than for Q other parameters determining filtration at or near levels observed before administration CsA. another group (N = 6) an identical dose given to measure circulating level endothelin. these rats, (measured radioimmunoassay) 41.7 ± 14.7 pg/ml, contrasting value
