Improved characteristics and protective efficacy in an animal model of E. coli-derived recombinant double-layered rotavirus virus-like particles

摘要: Live rotavirus vaccines that are effective in middle- and high-income countries have been found to be less immunogenic infants resource-limited settings. The virus-like particle (VLP) approach is promising for vaccine development, but challenges remain VLP production at large scale. In this study, capsid VP2 VP6 proteins were expressed Escherichia coli assembled with high efficiency into homogeneous single-layered VP6-VLPs or double-layered VP2/6-VLPs (dl2/6-VLPs) through a post-purification assembly process. dl2/6-VLPs observed better thermal stability antigenicity. Although the immunogenicity of trimers, was comparable, efficacy protect against rotavirus-induced diarrhea pups significantly higher than trimeric when assessed using mouse maternal antibody model. Taken together, recombinant dl2/6-VLP antigen, which highly analogous virion-derived particles, viable candidate development has potential parenterally administered safe efficacious vaccine.
