摘要: Vesicular Over-expressed in cancer Prosurvival Protein 1 (VOPP1), also known as Glioblastoma Amplified and Secreted EGFR-Coamplified has been previously shown to be over-expressed human glioblastoma multiforme squamous cell carcinoma. Additionally, previous experimental work suggests that it confers a prosurvival cellular phenotype. A query of public database gene expression profiling data (Oncomine™) shows the VOPP1 transcript is highly expressed several other common cancers, including breast carcinoma, pancreatic lymphoma. Analysis sequence structure both signal transmembrane domain, examination microarray dataset for endoplasmic reticulum (ER)-bound mRNA transcripts consistent with protein product being synthesized into ER. Immunoblot analysis culture conditioned media confirms not secreted retained intracellularly. tagged fluorescence reporter, well antibody-mediated visualization recombinant native forms reveals an intracellular vesicular pattern localization. Co-localization experiments reveal vesicles do co-localize mitochondria or peroxisomes, but show partial co-localization perinuclear lysosomes. markers endocytosis autophagy co-localization, suggesting VOPP1-containing enter final pathways lysosomal system. These findings throw doubt hypothesis interacts directly cytoplasmic mediators NF kappa B pathway, suggest phenotype conferred by this mediated mechanisms.
