Co-dependence between trypanosome nuclear lamina components in nuclear stability and control of gene expression

摘要: The nuclear lamina is a filamentous structure subtending the envelope and required for chromatin organization, transcriptional regulation maintaining structure. trypanosomatid coiled-coil NUP-1 protein component functionally analogous to lamins, major proteins of metazoa. There little evidence shared ancestry, suggesting presence distinct system in trypanosomes. To find additional components we identified interacting by affinity capture mass-spectrometry. Multiple pore complex (NPC) second protein, which termed NUP-2, were found. NUP-2 has punctate distribution at periphery throughout cell cycle close proximity NUP-1, NPCs telomeric chromosomal regions. RNAi-mediated silencing leads severe proliferation defects, gross alterations structure, organization architecture. Further, transcription altered telomere-proximal variant surface glycoprotein (VSG) expression sites (ESs), role controlling ES expression, although does not increase VSG switching. Transcriptome analysis suggests specific Pol I-dependent transcription. mislocalized knockdown cells vice versa, implying that form co-dependent network identifying as component.
