Effects of continuous ingestion of green tea or grape seed extracts on the pharmacokinetics of midazolam.

摘要: Limited systematic data on herb-drug interaction are available, despite many opportunities to concomitant use of herb with prescribed drugs. We investigated the effects 15 herbal extracts in dietary supplements CYP2C9, CYP2D6 and CYP3A4 activities human liver microsomes. Strong inhibition these CYP was found by addition green tea (GTE) or grape seed (GSE) vitro. To examine CYP3A vivo, pharmacokinetics midazolam (MDZ) analyzed rats. Although single treatments had negligible effects, 1 week treatment them resulted a significant increase ke intravenously administered MDZ, indicating induction liver. In contrast, GTE, but not GSE, caused C(max) AUC(0-infinity) orally MDZ without change t(1/2), suggesting reduction activity small intestines. These studies indicate that subchronic ingestion GTE GSE may alter appear opposite between intestine, which could be predicted from vitro experiments.
